Does Windows list all of your available serial ports? You can check available ports and assigned COM number in device manager under "Ports".
Did you run the DriveWire "Simple Config Wizard"? If you haven't, you can run it from the Config menu option.
Does DriveWire list the available ports?
Did you select the correct instance of the DriveWire server? If you haven't you can do so under "File-->Choose Instance"
